ABSTRACT

Objective To investigate the clinical effect of traditional Chinese medicine syndrome differentiation-based treatment combined with entecavir in the treatment of chronic hepatitis B. Methods A total of 80 outpatients with chronic hepatitis B were randomly divided into the observation group (n=40) and control group (n=40).The patients in the control group were treated with entecavir tablets. The patients in the observation group were given TCM syndrome differentiation-based treatment in addition to the treatment received by the control group. Clinical symptoms relief, improvement of liver function indexes, serological conversion and HBV-DNA negative rate were compared between the two groups after 48 weeks of treatment. Results The clinical symptoms of abdominal distension, fatigue, pain and anorexia were relieved better in observation group than in control groups. The difference was significant between the two groups (P<0.05). The observation group had significant therapeutic advantages over the ALT recurrence rate compared to the control group, especially 24 weeks ago (P<0.05). There was no statistically significant difference between the combined group and the control group after 12, 24, 48 weeks after treatment, and the HBV-DNA (both greater than 0.05) were clinically modified. Conclusion TCM syndrome differentiation-based treatment combined with entecavir had significant therapeutic advantages in the treatment of chronic hepatitis B, which could relieve clinical symptoms, improve liver function indexes, and converse serological changes and be worthy of clinical popularization.

ABSTRACT

Drug-receptor interaction plays an important role in a series of biological effects, such as cell pro-liferation, immune response, tumor metastasis, and drug delivery. Therefore, the research on drug-re-ceptor interaction is growing rapidly. The equilibrium dissociation constant (KD) is the basic parameter to evaluate the binding property of the drug-receptor. Thus, a variety of analytical methods have been established to determine the KD values, including radioligand binding assay, surface plasmon resonance method, fluorescence energy resonance transfer method, affinity chromatography, and isothermal ti-tration calorimetry. With the invention and innovation of new technology and analysis method, there is a deep exploration and comprehension about drug-receptor interaction. This review discusses the differ-ent methods of determining the KD values, and analyzes the applicability and the characteristic of each analytical method. Conclusively, the aim is to provide the guidance for researchers to utilize the most appropriate analytical tool to determine the KD values.

ABSTRACT

Objective:To develop an HPLC-MS determination method for the effects of Xuesaitong on the pharmacokinetics of losartan in rats. Methods:After oral administration of single losartan and combined usage of losartan and Xusaitong in rats,the con-centrations of losartan and EXP3174 in plasma were determined by HPLC-MS. The main pharmacokinetic parameters were calculated and compared by statistical analysis. Results:The calibration curves of losartan and EXP3174 were linear within the range of 2.0-2 000 ng·ml-1(r=0.996 4 and r=0.999 5). The average recovery was 85.19%-109.79%. The results showed that after oral co-ad-ministration with Xuesaitong,t1/2and Vz/F of losartan significantly decreased (P<0.05), Tmaxsignificantly increased (P<0.05), t1/2of EXP3174 significantly decreased(P<0.05),and Tmaxsignificantly increased(P<0.05). Conclusion:The HPLC-MS method is sensitive and selective,and suitable for the detection of losartan and EXP3174 in plasma in vivo. The experiments showed that Xue-saitong can affect the absorption and metabolism of losartan in vivo.

ABSTRACT

Objective To do screening acute lymphoblastic leukemia patients scFv antibody single chain variable region to cre-ate conditions for the expression and obtain further specificity of antibody fragments.Methods In this study,patients with newly diagnosed acute lymphoblastic leukemia serum as coating antigen using phage display technology,screening phage an-tibody specificity from the semi-synthetic human phage antibody libraries,the first to target the immune antigen-coated tab-let,phage library was added,so that with the target antigen-specific binding phage antibody was immobilized on plates immu-nization,could not be specifically bound phages were rinsed.The eluted specific binding phage,E.coli infection.Could get the specific antibody gene containing phagemid.Results After three “adsorption-elution-amplification”screening process,got stronger leukemia patient antigen-specific phage antibody variable region fragment and identification.Conclusion Got better strain affinity antibody fragments,to create the conditions for the next fragment expression,identification and clinical appli-cation.

ABSTRACT

Objective To screen permethrin human single-chain variable region (scFv) antibody for aims of developing rapid detection kit. Methods Phage display technology was used in this study. Permethrin was solid phase coated on Nunc plate as antigen. Semi-synthetic single-chain variable region of human antibody library technology was applied, and single chain variable region was screened from phage antibody library after 3 rounds "adsorption - elution - amplification" of the selection process. 100 clones were random selected as resistance to permethrin clones , enzyme-linked immunosorbent assay (ELISA), crossreactivity and competitive inhibition experiments were used to validate permethrin binding activity with strong scFv clones from the selected phage antibody clones plasmid. The plasmid was digested with restriction enzyme Sfi Ⅰ / Not Ⅰ and subcloned into pCANTAB5E vector. After transformed into E. coli XL1BIue, the plasmid was identified by restriction enzyme analysis. Results After screening in 100 clones, 18 clones had high ELISA absorbance values ( A value) at 490nm wavelength ( A490nm), then bovine serum albumin (BSA) cross-reactions identified five weak cross-reaction. Combined with the triplicate ELISA and competitive inhibition experiment results, one positive clone was acquired at last. And this clone was subcloned into pCANTAB5E vector and transformed into competent cells XL1-Blue. Conclusion Plasmid fragment was consistent with the purpose, which provided the foundation for further study of its specific affinity.

ABSTRACT

BACKGROUND: Parental rearing styles has profound and long-range effects on the mental development of their children.OBJECTIVE: To investigate the relationship between parental rearing styles and its correlative factors and the mental health of medical students.DESIGN: A transactional survey study based on medical students.SETTING: Department of psychology in a clinical medical college of a normal school.PARTICIPANTS: The study was conducted in the faculty of medicine of Hangzhou Normal School in December 2002. Totally 165 students of grade 2000-2002 including 106 college students, 59 undergraduates, and 98males, 67 females were selected as subjects. The youngest was 19 years old and the oldest was 24 years old. Totally 161 students had both parents (97.60%), 4 students had single parent (2.40%), 53 students were the only child of their family (32. 10%), and 112 students were not the only child of the family (67.90%).METHODS: Egna minnen barndoms uppfostran(EMBU) and symptom checklist 90(SCL-90) was introduced to evaluate the parental rearing styles and the mental health level in 165 medical students. SPSS 10.0 software was used for correlation analysis and multifactor stepwise regression analysis.MAIN OUTCOME MEASURES: Scores for 11 factors in EMBU including father' s emotional warmth and understanding, father' s punishment and stringency, father' s excessive interference, father' s favoritism, father' s rejection of deny, father' s excessive protection, mother' s emotional warmth and understanding, mother' s excessive interference, mother' s excessive protection, mother' s punishment and stringency, and mother' s favoritism, and the selection status of the factors correlated with the parental rearing styles including the relationship between father and mother, family income, and family type, etc. Total mean and scores for 9 factors in SCL-90 including somatization, obsessive-compulsive, interpersonal sensitivity, depression, anxiety, hostility, phobic anxiety, paraniod ideation and psychoticism.RESULTS: Correlation analysis displayed that there was significant positive correlation between father' s punishment and stringency and somatization,sensitive human relationship(r=0. 186, r=0. 162), between mother' s punishment and stringency and sensitive human relationship( r =0. 185),between father' s favoritism and obsessive-compulsive, interpersonal sensitivity, depression, anxiety, hostility, phobic anxiety, paraniod ideation and psychoticism factor and total mean( r = 0. 161, 0. 243, 0. 224, 0. 182,0. 166, 0. 193), between mother' s favoritism and depression( r=0.132),between father' s excessive protection and depression, anxiety, and total mean( r = 0. 177, 0. 204, 0. 162), and between mother's rejection of deny and sensitive human relationship( r = 0. 162) . There was significant negative correlation between father' s emotional warmth and anxiety, hostile ( r =-0. 187,-0. 241 ). As indicated by multifactor stepwise regression analysis for the impacts of parental rearing styles and its correlative factors on SCL-90 total mean, rearing styles including mother' s education background and father' s excessive protection entered into regression equation (β= -0. 269, P =0. 001; β=-0. 187, P=0. 016).CONCLUSION: Parents' punishment and stringency, parents' favoritism, father' s excessive protection and mother' s rejection of deny are not good for the mental health of medical students. Father' s emotional warmth is good for the improvement of mental health in medical students. The impacts of father' s excessive protection and mother' s education background are even more important to the mental health of medical students. Hence, the improvement of mental health in students should extend to the instruction of parental rearing styles.

ABSTRACT

<p><b>BACKGROUND</b>To investigate the immunohistochemical detected method and the clinical incidence of the bone marrow micrometastases (BMM) in patients with non-small cell lung cancer (NSCLC) and to analyze the sensitivity and specificity and clinical application value.</p><p><b>METHODS</b>Bone marrow samples were collected from the anterior superior iliac spines or posterior superior iliac spines of 53 patients with NSCLC in clinical stage I to III and 15 patients in stage IV, and the BMM was detected by immunohistochemical techniques (IHC) using monoclonal antibodies AE1/AE3 against cytokeratin. Chi-square test was used statistically.</p><p><b>RESULTS</b>The IHC sensitivity could be 10⁻⁵. The BMM positive rate was 22.6% (12/53) in stage I to III and 53.3% (8/15) in stage IV, and there was a significant difference in the BMM positive rate between stage I to III and stage IV (P < 0.05). No correlation was observed between BMM and sex, age, KPS, pathology classification and cancer cell differentiation.</p><p><b>CONCLUSIONS</b>The detection of BMM by IHC is convenient, sensitive, and specific. It might be helpful to diagnose bone marrow micrometastasis in patients with NSCLC.</p>

ABSTRACT

Objective To screen and identify the human phage engineered antibodies against HBsAg, and to analyze their gene sequence. Methods The phage antibody library was constructed by phage surface display techniques, and then the antibody genes were isolated and amplified from human peripheral blood lymphocyte. The human phage engineered antibody against HBsAg was selected through bio-panning from the phage antibody library. The affinity and specificity of the phage antibody was identified by ELISA and inhibition ELISA assay. The genes of heavy chain and light chain of the phage antibody against HBsAg were analyzed by sequencing. Results 30 colonies were obtained after three rounds of bio-panning. One of them had the highest A490 (A490=1.47?0.08) with ELISA, and the inhibition rate was 76% with inhibition assay (A490=0.35?0.10), implying that the phage antibody against HBsAg was of high specificity. The findings of enzyme digestion demonstrated that the phage antibody included genes of heavy chain and light chain. Sequencing results indicated that the VH region of heavy chain belonged to VHI subgroup and the VL region of light chain belonged to V? Ⅰ and V? Ⅲ subgroup. Conclusions The specific human phage engineered antibody against HBsAg was selected successfully from the phage antibody library. These results suggest that screening and identification of human phage engineered antibody against HBsAg through the phage antibody library are technologically feasible. It lays a foundation for application of human engineered antibody against HBsAg to design new targeted therapy strategy for HBV.
